Association of angiopoietin-like protein3 (ANGPTL3) with HTGL and LPL activities in human plasma

چکیده
Background: The relationship between plasma angiopoietin-like protein 3 (ANGPTL3), and lipoprotein lipase (LPL) activity and hepatic triglyceride lipase (HTGL) activity has not been investigated in the metabolism of remnant lipoproteins and HDL. Methods: Angiopoietin-like protein-3 (ANGPTL3), LPL activity, HTGL activity, remnant lipoproteins (RLP-C & RLP-TG), small dense LDL-Cholesterol (sd LDL-C) were measured in 20 overweight and obese subjects in the fasting and postprandial state. Results: Plasma TG, RLP-C, RLP-TG and sd LDL-C were inversely correlated with LPL activity both in the fasting and postprandial state, but not correlated with HTGL activity and ANGPTL3. However, plasma HDL-C was positively correlated with LPL activity both in the fasting and postprandial state, while inversely correlated with HTGL activity. ANGPTL3 was inversely correlated with HTGL activity both in the fasting and postprandial state, but not correlated with LPL activity. .Conclusion: Plasma remnant lipoproteins and sdLDL-C were inversely correlated with LPL activity but not correlated with HTGL activity and ANGPTL3, indicating that LPL, but not HTGL, plays a major role in the metabolism of remnant lipoproteins including small dense LDL. However, HDL-C was inversely correlated with HTGL activity and positively correlated with ANGPTL3, indicating that HTGL plays a major role in HDL metabolism but not remnant lipoprotein metabolism. These findings suggest that ANGPTL3 is strongly associated with the inhibition of HTGL activity and regulates HDL metabolism, but not associated with the inhibition of LPL activity for the metabolism of remnant lipoproteins in human plasma..

Angiopoietin-like protein3 regulates plasma HDL cholesterol through suppression of endothelial lipase.
OBJECTIVE A low level of high-density lipoprotein (HDL) in plasma has been recognized as an aspect of metabolic syndrome and as a crucial risk factor of cardiovascular events. However, the physiological regulation of plasma HDL levels has not been completely defined. Current studies aim to reveal the contribution of angiopoietin-like protein3 (angptl3), previously known as a plasma suppressor o...
متن کاملA decreased expression of angiopoietin-like 3 is protective against atherosclerosis in apoE-deficient mice.
KK/Snk mice (previously KK/San) possessing a recessive mutation (hypl) of the angiopoietin-like 3 (Angptl3) gene homozygously exhibit a marked reduction of VLDL due to the decreased Angptl3 expression. Recently, we proposed that Angptl3 is a new class of lipid metabolism modulator regulating VLDL triglyceride (TG) levels through the inhibition of lipoprotein lipase (LPL) activity. In this study...
متن کاملSpecies, diet, and gender differences in plasma postheparin lipolytic activities in nonhuman primates. Relationships with plasma lipids and high density lipoproteins.
Studies were done in selected groups of nonhuman primates in which the diet (low fat and high fat as either polyunsaturated or saturated fat), gender, and species (African green monkeys and cynomolgus monkeys) were varied. Large differences in plasma lipid and lipoprotein concentrations were found among these groups, providing the opportunity to examine their relationships to postheparin lipoly...
متن کاملRegulation of lipid metabolism by angiopoietin-like proteins.
PURPOSE OF REVIEW The angiopoietin-like proteins (ANGPTLs) 3, 4 and 8 have emerged as key regulators of plasma lipid metabolism by serving as potent inhibitors of the enzyme lipoprotein lipase (LPL). In this review, we provide an integrated picture of the role of ANGPTL3, ANGPTL4 and ANGPTL8 in lipid metabolism by focusing on their impact on LPL activity and plasma triglyceride clearance during...
متن کاملANGPTL8 promotes the ability of ANGPTL3 to bind and inhibit lipoprotein lipase
OBJECTIVE Several members of the angiopoietin-like (ANGPTL) family of proteins, including ANGPTL3 and ANGPTL8, regulate lipoprotein lipase (LPL) activity. Deficiency in either ANGPTL3 or ANGPTL8 reduces plasma triglyceride levels and increases LPL activity, whereas overexpression of either protein does the opposite. Recent studies suggest that ANGPTL8 may functionally interact with ANGPTL3 to a...
